How Do We Validate Our Knockout Antibodies?
A three-step process built around a real genetic knockout, not a synthetic blocking control.
Generate the knockout
We create CRISPR knockout cell lines by introducing frame-shift mutations into the genomic DNA of target cells using sgRNA/Cas9.
Run a parallel Western blot
The CRISPR knockout cells are lysed alongside wild-type (WT) control cells and run side-by-side on a Western blot.
Confirm loss of signal
Blots are probed with our antibody. A clean loss of signal in the knockout lane, against a persistent WT band, confirms high specificity.

What Is KO Validation?
KO validation is a means by which antibody specificity is confirmed by testing antibodies in a knockout (KO) cell line. Using KO cell lines — cells with no expression of the target protein — is the new gold standard for validating antibody specificity.
If a signal is detected from a KO cell line, the antibody is not highly specific; this is due to the antibody binding to a similar protein within the sample. Antibodies with high specificity, on the other hand, will not produce any signal in the knockout. KO validated antibodies must be shown to be specific, selective and reproducible.
Why Is Validation Important?
Antibodies are commonly used research tools, applied routinely in experiments such as Western blot, ELISA, immunostaining (IHC/ICC), immunofluorescence and flow cytometry. For the findings of these techniques to be reliable, they must have an internally high standard of accuracy and precision.
This is why there is a need for highly specific antibodies that can ensure reproducibility of results — and why every antibody in this range is put through the KO validation process described above before it's listed for sale.

What does “KO Validated” mean on an antibody datasheet?
It means the antibody has been tested side-by-side on a CRISPR/Cas9 knockout (KO) cell line and its wild-type (WT) counterpart by Western blot. If the signal disappears in the KO sample while the WT band remains, the antibody is confirmed to bind specifically to the intended target rather than a similar off-target protein.

Why is KO validation considered the gold standard for antibody specificity?
Peptide-blocking and overexpression assays can still leave cross-reactivity with related family members undetected. A true genetic knockout removes the target protein entirely, so any signal that remains can only be non-specific binding — making it the most rigorous specificity test currently available for research antibodies.
Which applications are KO validated antibodies suitable for?
Every antibody in this range is validated by Western blot (WB). Many are additionally tested and recommended for ELISA, immunohistochemistry (IHC), immunofluorescence/ICC, immunoprecipitation (IP), flow cytometry (FC) or ChIP — check the Application field on each individual product page for that specific antibody.
Can I use a KO validated antibody in a species that wasn’t part of the knockout study?
Reactivity is listed on each product page (for example Human, Mouse, Rat). Knockout validation is typically performed in one species/cell line, but sequence homology across the listed reactive species means the antibody is generally reliable in those species too — contact our technical team if you'd like help confirming cross-reactivity for your specific model.
